生物多样性与群落稳定性的关系一直以来是土壤生态学研究的热点问题之一[1]。一般认为,食物网结构越复杂,生物多样性越高,生态系统的稳定性越强。这是由于复杂的食物网中某一生物类群消失,可由相似功能的生物代替,从而可以保持生态系统的稳定[2],相比之下简单食物网某一食物链的断裂则可能对生态系统功能产生较大影响。在陆地生态系统中,土壤样品易于采集,土壤生物的提取和鉴别技术日益成熟,为研究生物多样性和生态系统功能稳定性的关系提供了良好平台。此前,已有不少学者开展了相关工作[3-6],然而,大部分研究集中在地下生态系统分解者的层面上[1,7],无法反映食物网的全貌,这样的缺陷使得研究对象转向能够提供更为丰富的土壤生态功能信息的指示性生物[8]。
在众多土壤生物中,线虫的种类丰富,数量庞大,广泛分布于地球上所有的土壤类型[9],其占据碎屑食物网的多个营养级[10],对土壤有机质的降解和养分循环起到重要作用[11]。依据世代周期和体型,线虫可划分成不同的生存对策者[12],cp值为1的r对策者可对土壤有机物投入做出迅速响应;世代周期略长的一般机会主义者(general opportunists)通常具有更强的扰动耐受性;cp3-5的偏K对策线虫则对干扰较为敏感,数量会减少甚至消失[12]。基于这些特性,线虫群落结构可用于指示土壤环境状况[13-14],并由此发展出了一系列用于反映土壤食物网结构和功能的生态指数和评价工具[12,15-16],广泛应用于分析各类扰动对土壤环境的影响[17-19]。
重金属污染是一类典型的土壤扰动,污染物通过大气沉降、污水灌溉、固体废弃物等途径进入土壤,威胁土壤生物的生存繁衍,影响地下生态系统的正常运转。单就线虫而言,已有大量相关报道[20],例如沿着污染梯度进行野外采样,探讨重金属污染程度与线虫群落的相关性[21-22];或者定性或定量控制试验,人为设置污染后观察线虫群落的变化[23-25]。Nagy[20]归纳具有明显影响的重金属元素包括Zn,Cu,Ni,Pb,Cr和Se。鉴于此,本试验选取代表性元素Cu和Zn为扰动因子,探索两种不同土壤中线虫群落对重金属污染的响应。所选样地为临近的同一土壤背景下的自然林地和农田,自然林地土壤具有更高的生物多样性,视为健康土壤;而农田由于长期的扰动,食物网相对简单,视为受胁迫土壤。有别于此前的报导[3,6],本研究将从污染程度的角度出发,明确健康土壤和胁迫土壤对重金属污染的抵抗力的差异,为后续从时间角度研究多样性和稳定性关系提供参考。
1 材料与方法 1.1 研究区概况土壤样品采集于北京市延庆区小丰营村(N40°26′0″,E115°53′2″),该区域属温带与中温带、半干旱与半湿润带的过渡地带,大陆性季风气候,冬冷夏凉,年均气温8 ℃。多年平均降水量443.2 mm,降水时空分布不均。区域内土壤类型为潮褐土,质地为轻壤。取样分别在临近的自然林地和农田进行。自然林地面积约824 m2,为多年荒地植被自然演替而成,几乎无人为干扰,其主要覆盖植被包括猪毛菜(Salsola collina)、刺儿菜(Cirsium arvense var. integrifolium)、艾蒿(Artemisia argyi)、苣荬菜(Sonchus wightianus)和刺儿槐(Robinia pseudoacacia)。农田位于自然林地北侧约43 m远,面积约1 452 m2,种植西兰花(Brassica oleracea)(取样期间已收获),并常年施用有机肥(25 t·hm-2)、复合肥(1.15 t·hm-2)和尿素(0.53 t·hm-2),人为扰动强烈。样地土壤基础理化性状见表 1。
土壤样品采集时间为2017年6月21日。为尽量降低土壤异质性,每种样地分别选择地面均一的1 m2作为采样样方,用木铲挖取0~20 cm土壤各20 kg,迅速带回实验室,常温保存,并于三日内完成培养布置工作。
每种土壤分别添加CuSO4和ZnSO4作为重金属扰动因子,根据参考文献[26]的添加量以及我国土壤Cu和Zn三级污染的标准,设计对照(CK)、低浓度(L)、中等浓度(M)和高浓度(H)的添加梯度,分别为0、100、400、800 mg·kg-1干土的添加量,其中CK为两种重金属共用,因此试验总计(1+3×2)×2=14个处理,重复3次。每盆装入310 g过4 mm筛的鲜土,根据土壤含水率换算出干土所需的重金属添加量。每盆自然林地和农田土壤分别加入80 mL和35 mL配置好的溶液,使得土壤达到田间持水量的60%。所有培养盆转移至25 ℃培养箱中避光培养,每周补充水分一次,同时随机调换位置;参考Liu [6]和Korthals [25]等的培养周期,于23 d后进行破坏性取样。
土壤基础理化性质的测定采用常规方法。土壤有效铜和有效锌含量采用0.1 mol·L-1盐酸浸提-原子吸收光谱法测定。Cu处理的土壤仅测定有效铜含量,Zn处理的土壤仅测定有效锌含量,而两种生境的对照处理有效铜和有效锌均测定。
1.3 线虫分离与鉴定线虫分离采用浅盘法。将60 g鲜土平铺于1 mm网筛上,并在网筛上垫一层纸巾,48 h后收集浅盘中的线虫,静置1 d后保存于4%福尔马林中。所获线虫于40倍倒置显微镜下进行计数,随机挑选100条于100-400倍显微镜下根据线虫头部、尾部、食道等特征鉴定至属水平。根据线虫网站(http://nemaplex.ucdavis.edu/Ecology/EcologyMenu.htm)上的食性分类,所获线虫划分为4个营养类群:食细菌线虫(Ba)、食真菌线虫(Fu)、植食性线虫(He)和杂食捕食性线虫(OP)。
1.4 数据处理通过NINJA系统(http://sieriebriennikov.shinyapps.io/ninja/)将线虫群落数据上传,以计算用于指示食物网状况和扰动程度的功能性生态指数,本试验中用到的指标包括富集指数EI(Enrichment index)、结构指数SI(Structure index)和基础指数BI(Basal index)。此外,还计算3个多样性指标:香农指数H′(Shannon index)、优势度指数λ(Dominance)和营养类群多样性指数T(Trophic diversity)。H′=-∑pi(lnpi),λ=∑pi2,其中pi为第i个分类单元个体数在线虫总数中的比例;T=1/∑pi2,pi为第i个营养类群在线虫群落中的比例[27]。
重金属扰动对线虫的影响采用单因素方差分析,多重比较方法为LSD,对于不满足齐次性检测的数据,方差分析前进行ln(x+1),开平方或反正弦转换,若转换后仍不满足齐次性要求,采取Kruskal-Wallis非参数检验,并用Mann-Whitney非参数检验两两比较处理间的差异。土壤之间,以及重金属之间的对比采用t检验。所有分析显著性水平设为P < 0.05,SPSS16.0中完成。
排序分析用来呈现不同处理间线虫群落的差异,采用PCA模型,并将数据进行ln(x+1)转化。分析在Canoco 5.0中完成。
2 结果 2.1 不同处理土壤有效Cu和Zn含量随着添加量的升高,自然林地和农田土壤中有效态Cu或Zn均显著升高(P < 0.05),有效铜最高值均出现在自然林地(363.79 mg·kg-1),有效Zn最高值出现在农田(297.27 mg·kg-1)(表 2)。
重金属施用梯度对线虫总数有显著影响,其中对自然林地的影响要强于农田土壤(图 1)。自然林地线虫数量随扰动增加而逐渐下降,并且在Zn的处理中更为明显;而在农田则呈现低浓度条件下线虫数量降低,中、高浓度下又升高的趋势。在中、高浓度处理中农田线虫总数高于自然林地,仅在Cu的中等浓度处理未见显著差异(P > 0.05);而在低浓度条件下,呈相反趋势,并且在Zn处理中差异显著(P < 0.05)。
自然林地中的植食性线虫数量显著高于农田土壤(P < 0.05)(图 1),且重金属对自然林地植食性线虫数量没有显著影响(P > 0.05),但对农田的表现出抑制趋势。食细菌线虫在各处理间的变化规律与线虫总数的相近。重金属对食真菌线虫总体上无显著影响(P > 0.05),仅在Zn处理的低浓度和高浓度之间表现出显著差异(P < 0.05)。对于捕食杂食性线虫,在自然林地仅添加高浓度Zn表现出显著抑制效应(P < 0.05);而在农田土壤,中、高浓度Cu,以及低浓度Zn均可显著降低线虫数量(P < 0.05)。
2.3 Cu和Zn添加对线虫群落结构的影响试验总计发现52个线虫分类单位,其中自然林地48个,农田32个,其中自然林地特有的线虫种类远多于农田的(表 3)。两种生境均以食细菌线虫为优势类群;植物寄生类线虫在自然林地为次优势类群,但其在农田土壤中所占比例最小。在自然林地中,重金属表现出抑制食细菌线虫比例,提升植食性线虫比例的规律;而在农田土壤则呈现相反的趋势(表 4)。
农田土壤中以Ba1线虫和Ba2线虫为主,且二者数量相当(表 5);自然林地中Ba2线虫占绝对优势,其次是Ba3线虫,且这两类线虫在中、高扰动条件下大幅减少。He3是自然林地主要的植食性线虫,其数量在高浓度扰动条件下有较明显的下降。杂食捕食性线虫以Om4为主,此外在自然林地中还存在相当比例的Pr5线虫,其数量随扰动程度的增加而减少。
线虫群落PCA分析显示,自然林地各重金属处理的线虫群落与CK相似度较高(图 2a),仅在中浓度的Cu和Zn,以及高浓度的Zn表现出和对照较大的差异;相比而言,农田土壤CK与各重金属处理后的线虫群落差异更为明显(图 2b),反映出添加重金属对农田土壤线虫影响更强烈。
重金属梯度对自然林地的优势度和香农指数无显著影响(P > 0.05)(图 3),但是显著提升了中、高浓度处理的营养类群多样性。对于农田土壤,添加重金属表现出降低线虫多样性,提高优势度的趋势,Cu处理中在高浓度时达到显著性差异(P < 0.05);Zn处理中低浓度时即表现出显著性差异(P < 0.05)。高浓度Cu添加显著降低了农田土壤线虫营养类群多样性(P < 0.05);而对于Zn组处理,低、中浓度表现出显著的降低效应(P < 0.05)。无论是对照,还是添加重金属后,自然林地线虫多样性(香农指数和营养类群多样性)均高于农田土壤,且除低浓度的Zn处理外均达到显著性差异(P < 0.05)。而优势度指数呈现相反趋势,但在对照、中浓度Cu处理中差异并不显著(P > 0.05)。
Cu对线虫功能性指数的影响要弱于Zn的影响(图 4)。Cu仅显著降低了农田土壤高浓度处理的结构指数(P < 0.05)。Zn添加对自然林地的富集指数无显著影响(P > 0.05),但对基础指数产生了先抑制,后提升的效应,并在高浓度条件下显著降低了结构指数(P < 0.05)。Zn对农田土壤各线虫功能性指数均有不同程度的影响,基础指数和富集指数呈现出波动状态,结构指数降低,并在中等添加浓度条件下呈显著差异(P < 0.05)。总体上,重金属添加梯度中自然林地线虫的结构指数高于农田土壤的,并且在大部分对应的试验组达到显著性差异(P < 0.05),而富集指数呈现相反的趋势,反映出自然林地食物网更为复杂,扰动较小,农田土壤呈养分富集状态,扰动强烈(图 4)。基础指数也表现为自然林地的高于农田的,但大部分对应组差异不显著(P > 0.05)。
在添加Cu扰动处理中,自然林地的有效Cu含量同线虫总数呈显著负相关关系(P < 0.05)(表 6),而同营养类群多样性指数呈显著正相关关系(P < 0.05);农田中有效Cu含量同结构指数和香农指数呈极显著负相关关系(P < 0.01),同线虫优势度呈显著正相关关系(P < 0.05)。在添加Zn扰动的处理中,自然林地的有效Zn含量分别同结构指数和线虫总数呈显著(P < 0.05)和极显著(P < 0.01)负相关关系;同营养类群多样指数成极显著正相关关系(P < 0.01);农田的有效Zn含量同线虫各指标均无显著相关性。综合了两种生境(n=24)的各线虫指标同有效Cu和Zn均无显著相关性。
土壤线虫是一类对环境污染和人为扰动反应敏锐的生物[12-13],通常在重金属浓度升高的土壤中,数量呈下降趋势[25,28- 29],本研究中自然林地线虫的数量的变化即符合这个规律。但是在农田土壤中,线虫数量出现低浓度污染条件下降低,中、高浓度条件下又反弹的现象。由于食细菌线虫在两种土壤中均是优势类群,因此线虫总数的表现与食细菌线虫密切相关。两种土壤中食细菌线虫群落结构本身存在明显差异,自然林地中以Ba2线虫为主,其次为Ba3线虫,这两类线虫在中和高Zn和Cu胁迫条件下均不同程度的减少,也就导致了食细菌线虫丰度的降低。而农田中世代周期较短、可以快速繁殖的Ba1线虫平均占到48.5%的比例,Clarke和Shepherd[30]曾报道了一定浓度的Cu和Zn能刺激线虫卵的孵化,增加线虫丰度,因而本研究中农田土壤中线虫数量的表现一定程度上与Cu或Zn的添加促进典型机会主义者繁殖有关。值得注意的是,农田中也有较高比例的Ba2线虫,但其变化趋势有别于自然林地的该类线虫。即便属于同一功能群,不同种类的线虫对等同扰动的响应也可能存在差别[31],从而影响两种生境食细菌线虫数量的变化趋势。在自然林地中,植食性线虫是次优势营养类群,其数量未受Cu和Zn添加的影响。植食性线虫丰富与其寄主密切相关,而自然林地植被覆盖度较高,土壤中植物根及其碎屑丰富,短时间内不易分解。由此可以推测,对于自然林地土壤中植食性线虫,食物资源相比Cu和Zn扰动是更为重要的影响因素。反观在农田土壤中,植食性线虫种类和数量本身稀少,且植物根系生物量小(根据地上植被情况推测,未实时测定),因而外源扰动因素的作用更为明显。
杂食捕食性线虫是线虫中的高级类群,其在线虫群落中的比例及由其发展出的结构指数常用来指示土壤食物网的复杂性[10,12]。在本研究中,自然林地相比农田土壤具有更为复杂的食物网结构,这点与Li等[27]报道的自然林地、农田和温室等土地利用方式下线虫群落特征的对比研究结果一致。值得注意的是,在施加同等条件的重金属Cu和Zn扰动后,农田土壤的杂食捕食性线虫数量相比自然林地受到的影响更为明显,其在低、中等浓度扰动条件下即显著降低,而自然林地土壤中,此类线虫仅在经历了高浓度胁迫才表现出显著的减少,反映出从污染程度的角度出发,自然林地土壤食物网结构具有更强的抵抗力。然而,此现象与高cp值线虫对扰动更为敏感[12]这个主流观点看似并不完全相符。事实上,扰动并不一定对所有高级线虫类群都造成负面影响,比如耕作和施肥扰动反而会促进Mylonchulus(Pr4)的发生[13];Zhao和Neher[31]发现Aporcelaimellus(Om5)和土壤Zn含量呈正相关趋势;Liu等[6]对比了不同扰动条件下两种土壤线虫群落的响应,发现干燥胁迫提高了退化土壤的线虫结构指数。本研究中杂食捕食性线虫对Cu或Zn扰动可能存在不同的响应表现。
自然林地中线虫的丰富度(分类单位数量)、香农指数和营养类群多样性均高于农田土壤,而优势度指数呈现相反趋势。加入重金属扰动后,自然林地线虫香农指数依然能保持较高水平,而在农田土壤中多样性显著降低。在一个相似的研究中,Carrascosa等[3]报道了自然林地线虫丰富度、食物网复杂性等指标在经历农药扰动后相比农田土壤表现出更强的抵抗力。尽管该研究是从时间的角度探索线虫群落的变化,且施加的扰动有别于本研究,但也反映了低扰动、自然的、食物网复杂的土壤更有利于维持生物多样性和群落稳定性[2]。自然林地的这种优势可能与多种因素相关。首先,在自然的生态系统中,丰富的有机质可以对土壤动物及其功能起到保护作用[32];此外,本研究中自然林地的土壤有效磷含量比农田低96.8%(表 1),而Wakelin等[33]发现低磷环境有利于维持土壤功能的抵抗力;最后,根据生物多样性保险假说[34],自然林地的群落多样性高,可能具备更多适应某种胁迫的互补型分类单位[35]。
相比线虫数量和生态指标,PCA图能更为直观的反映出不同处理线虫群落的变化。很显然,在相同的扰动条件下,农田土壤线虫群落的变化比自然林地明显,这也与线虫生态指数的结果一致。PCA分析还显示在自然林地中,Zn扰动对线虫群落的影响大于Cu的扰动,但与Korthals等[25]报道的等同Cu和Zn对线虫群落的影响相似这个规律并不完全相符。
4 结论两种生境的土壤在经历Cu和Zn扰动后线虫数量和群落结构表现出不同的响应趋势。自然林地土壤线虫数量随Cu和Zn的添加浓度升高而降低;而农田土壤线虫呈现低浓度扰动条件下降低,中、高浓度又反弹的现象。且相比自然林地,Zn和Cu扰动对农田土壤线虫群落结构、多样性和生态指数影响更为明显。因而,从污染程度以及生物群落结构的角度来看,高多样性的自然林地对Cu和Zn污染的抵抗力更强。未来将从时间的角度继续探索线虫群落的稳定性,为阐明土壤生物多样性与稳定性关系提供佐证。
