摘要:利用功能细菌辅助植物固定重金属是目前农田土壤污染修复中高效且环境友好的方式,其中细菌与矿物间相互作用广泛存在,包括细菌对矿物的溶解作用、矿物对细菌活性的影响以及细菌-矿物复合体的形成等,并贯穿整个修复过程。一方面,细菌与矿物互作会影响细菌的活性和表面特性,如带电性、表面官能团位点类型及浓度等,进而影响细菌对重金属的生物吸附行为以及辅助植物修复作用的发挥;另一方面,细菌-矿物结合形成的复合体较单一细菌、矿物组分对重金属的固定行为不同,在重金属修复过程中发挥不可忽视的作用。本文综合分析细菌与矿物的结合作用、细菌对矿物的溶解作用以及矿物对细菌活性的影响,阐述细菌-土壤矿物(矿物材料)复合体在重金属污染修复中的应用潜能,为复合体应用于重金属污染土壤环境提供理论依据。

Interaction Between Bacteria and Soil Minerals and Application of Bacterial-Mineral Composites in Remediation of Heavy Metals Polluted Soil

Abstract: The use of functional bacteria to assist phytostabilization is nowadays a practical and eco-friendly method commonly applied to remediation of heavy metals-contaminated farmland soils. The application can effectively control the migration of heavy metal pollutants, reduce the concentration of available heavy metals in the soil, and further on ensure safety of the agricultural production. However, bacteria do not exist independently in soil. Nearly 80%~90% of the microorganisms exist on and adhere to the surface of soil minerals or mineral-organic complexes. Within this in-situ soil remediation process, the interplay between bacteria and minerals, including the formation of bacterial-mineral complexes, the dissolution of minerals by bacteria metabolites and the influence of minerals on bacterial activity, which runs through the whole remediation, should be taken in consideration and show extensive interferences. First, the interplay will affect the surface characteristics of bacteria, such as chargeability, type and concentration of the surface functional groups, etc., which in turn affect biosorption behavior of the bacteria to heavy metals; Secondly, minerals can affect bacterial activity by destroying integrity of the bacterial biofilm, dissolving out mineral ions to poison bacteria and buffering pH in the environment, etc., and disrupt internal physiological regulation mechanisms of the bacteria, which ultimately affect colonization ability of the bacteria on the surface of minerals, growth-promoting function of the bacteria on plants, and ability of the bacteria to immobilize heavy metals. Moreover, the composites formed by combination of bacteria and minerals differ in behavior in immobilization of heavy metals from a single component of bacteria or minerals. This process might promote formation of soil aggregates, improve soil physical structure, and slow down soil degradation, which is conducive to the sustainable use of remedied farmlands, and hence plays an important role in remediation of heavy metals-contaminated soils. At present, the research on relationships between soil minerals, bacteria and heavy metals is still limited, and most of them focus on apparent phenomena. Therefore, this paper is oriented to comprehensively review the combination of bacteria and minerals, the dissolution effect of bacteria on minerals and the influence of minerals on bacterial activity, and expound the application potential of bacteria-soil mineral(mineral material)complexes in remediation of heavy metal polluted soils, so as to provide a theoretical basis for the application of bacteria-mineral complexes in the heavy metals -contaminated soil environment.

利用功能细菌辅助植物修复是当前重金属修复的研究热点。一些重金属耐性细菌可分泌植物促生物质,如植物生长素、铁载体、1-氨基环丙烷-1-羧基(1-aminocyclopropane-1-carboxylate,ACC)脱氨酶等促进植物生长。此外,除促生作用外,重金属耐性细菌还可以通过分泌螯合物质、氧化还原反应等改变土壤中重金属的形态和有效性。针对不同的植物修复技术,细菌辅助植物修复的策略不同[1]:一方面,细菌可分泌有机酸、铁载体等活化重金属,提高植物提取过程中重金属转移率(植物提取)。Dimkpa等[2]报道重金属胁迫下Streptomyces tendae F4产生的铁载体显著提高了豇豆植物对Cd的吸收。另一方面,细菌表面官能团及胞外分泌物等可固定重金属[3],或通过氧化还原作用转化重金属,降低土壤中重金属的生物有效性,辅助植物根际固定或阻隔重金属(植物稳定)。Joshi和Juwarkar[4]发现,Azotobacter spp.的胞外聚合物(extracellular polymeric substance,EPS)能固定土壤中的Cd和Cr,从而降低Triticum aestivum的吸收量。两种修复策略中,针对农田重金属污染土壤环境,重金属耐性细菌辅助植物稳定这一策略被广泛应用。这一措施能有效控制重金属污染迁移、降低土壤中有效态重金属浓度,进一步保障农业生产安全。

目前,重金属耐性细菌辅助修复重金属污染土壤的研究主要集中在植物与细菌的相互作用[5-6],包括细菌如何促进植物生长,细菌如何固定重金属[7]。然而,细菌在土壤中并不是独立游离存在的,近80%~90%的微生物最终附着在土壤矿物或矿物-有机物复合体上形成细菌-矿物复合体。细菌与矿物相互作用(如图 1)一方面涉及两者电位、表面位点密度的改变,从而影响细菌对重金属的积累作用;另一方面,矿物会对细菌的活性造成影响,扰乱其内在生理调控机制,最终影响细菌在矿物表面的定殖能力、细菌对植物的促生作用以及细菌对重金属的固定能力。因此,本文综合分析土壤矿物与细菌的相互作用,包括细菌与矿物的结合作用、细菌对矿物的溶解作用、矿物对细菌活性的影响,阐述细菌-土壤矿物(矿物材料)复合体在重金属污染修复中的应用潜能。

1 细菌与矿物的相互作用

土壤环境中细菌广泛存在,大部分细菌黏附于矿物表面,与矿物结合并发生一系列相互作用,包括细菌对矿物的溶解、矿物对细菌活性的影响等,相关研究总结见表 1。细菌可通过分泌有机酸、无机酸等溶解矿物,而矿物可对细菌的活性造成影响(如溶出的矿质离子会对微生物产生毒性等),进而影响细菌的代谢过程。这一系列相互作用可改变矿物与细菌的表面结构、表面电荷以及官能团浓度与类型等一系列表面特性,最终对土壤中重金属的环境行为造成影响[8]

1.1 细菌与矿物的结合

土壤矿物与细菌的结合是一个复杂的动态反应,一般包括以下几个过程(如图 2):(1)初始附着:细菌通过鞭毛运动、趋化性等逐渐接近矿物界面,与矿物接触并黏附于矿物表面,这一过程是可逆的;(2)紧密黏附:细菌与矿物表面接触后改变自身生理生化性质或分泌脂质、多糖等物质紧密结合在矿物表面,这一过程不可逆;(3)形成微菌落或生物膜:细菌在这一过程中分泌EPS并进一步形成黏胶层,被胶结的细菌最终定殖在矿物表面发展成为生物膜。生物膜对于矿物与细菌的结合过程非常重要,它使两者间的结合更为紧密,同时也是细菌抵御外界不良环境的重要屏障。其中,EPS被证实在生物膜形成过程中扮演着重要角色[27],并且微菌落细胞的群体感应被认为是调控EPS产生和微生物膜形成的重要途径之一[28]。Zhang等[9]研究了Acidianus sp. DSM 29099在黄铁矿和黄铜矿表面的初始附着及生物膜的形成过程,发现分别有60%和35%的细菌在2 h内黏附于两种矿物表面,2~4 d后可在黄铁矿表面形成成熟的生物膜。

矿物与细菌的结合通常是在范德华力、静电力、氢键等多种作用力共同作用下发生的[29]。传统的Derjaguin-Landau-Verwey-Overbeek(DLVO)理论认为,细菌与矿物的结合是范德华力和静电作用力的平衡结果[30-31],而扩展的DLVO理论则认为细菌与矿物的结合是范德华力、静电作用力以及疏水作用力三者共同作用的结果[32]。此外,细菌表面羧基、羟基、磷酸基团与矿物表面羟基基团能够特异结合形成细菌-矿物复合体。Ren等[33]发现在复合体形成过程中,枯草芽孢杆菌的羟基、磷酸基团与三石铝石表面的羟基相结合。Hong等[34]的研究也发现枯草芽孢杆菌上的磷酸基团可与氧化铁矿物上的羟基发生配位体交换,并与铁原子配位形成Fe-O-P化学键。这一过程可利用傅里叶变换红外光谱技术进行分析验证,通过分析图谱中的波峰可得到细菌与矿物表面的官能团情况,通过吸附前后波峰的位移情况可分析得出两表面参与结合的基团。

细菌与矿物的吸附最初发生在细菌的细胞壁和矿物表面,当前研究主要通过吸附动力学和吸附热力学等方法拟合矿物与细菌的初始吸附行为。在吸附动力学研究中,准一级动力学模型(Pseudo-first-order model)和准二级动力学模型[35](Pseudo-second-order model)是常用于分析吸附量随时间变化情况的两种模型。其中,准一级动力学模型假定吸附随时间变化的速率和吸附最大量与吸附剂随时间的吸附量之间的差值成比例,而准二级动力学模型则假定吸附是受化学吸附机理控制进行的[35]。等温吸附研究则通常使用Langmuir和Freundlich两种模型进行拟合[36]。其中,Langmuir等温吸附模型认为吸附是吸附质发生在吸附剂表面的单层吸附[12, 37],而Freundlich等温吸附模型则是基于吸附质在多项表面上的吸附[38]。此外,影响初始吸附效果的因素主要分为两方面,一方面是矿物和细菌本身的特性,包括矿物的类型和表面性质、细菌的种类等,其中表面积越大、带正电荷越多的矿物越有利于对细菌的吸附[10, 39]。Chen等[10]研究了Acidithiobacillus ferrooxidansAcidithiobacillus caldus在黄铜矿表面的吸附行为,发现相同初始浓度下Acidithiobacillus ferrooxidans与黄铜矿的结合更快、结合量更大。Jiang等[39]报道,由于矿物类型和表面性质的不同,假单胞菌更倾向于吸附在带正电的针铁矿表面,其次为高岭石和蒙脱石。另一方面是环境条件的影响,如pH、离子强度等。随着溶液中pH的提高,矿物与细菌表面去质子化,矿物表面所带正电荷减少,对细菌的吸附量降低。而离子强度通过影响带电双电层进而对吸附造成影响。Yee等[13]发现刚玉对枯草芽孢杆菌Bacillus subtilis的吸附量随pH值增加而减少,并且随着离子强度增大,其吸附量也相应减小。Zhang等[40]报道,随着pH从10.0降至3.0,大肠杆菌O157:H7在高岭石和针铁矿表面的附着量增加。Rong等[41]发现随着电解质浓度从1 mmol·L-1增加至100 mmol·L-1,假单胞菌在针铁矿表面的黏附力增加11%。与此相反,Cai等[20]却观察到,离子强度的增加导致大肠杆菌O157:H7在针铁矿上的沉积量减少。这种差异可能与不同离子类型造成的效应不同有关。此外,矿物与细菌的吸附量随着细菌浓度、反应时间的增加呈现出先增长后稳定的趋势。

1.2 细菌对矿物的溶解作用

细菌对矿物的溶解作用一般包括两种方式。一是质子交换作用:溶液pH在质子交换过程中起决定性作用。细菌通过分泌有机酸、无机酸等改变矿物pH环境促使矿物溶解,释放矿质离子[42]。Ehrlich[14]的研究认为有机或无机酸、多聚糖等以及微生物表面电离出的H+均能与长石中的Na+、K+、Ca2+等发生质子交换,从而促进长石溶解。二是络合作用:细菌作用形成的阴离子配体与矿物中的Si、Al、Ca、Mg、Fe等离子络合,使之以络合物的形式从矿物的晶格中脱离,促使矿物溶解。参与络合作用的配体通常包括细菌代谢产生的低分子量有机酸、多聚糖及胞外多糖等。Casey和Ludwig[43]研究报道,有机酸中的羟基和羧基易与Si、Al、Fe等形成络合物。Welch和Ullman [44]研究发现在相同pH条件下,草酸较硝酸溶解硅酸盐的速率更快,因为有机酸不仅能发生质子交换,也可通过络合作用溶解矿物。此外,胞外聚合物、铁载体以及生物膜的形成同样对矿物的溶解存在促进作用。Torres等[16]以(Mg,Fe)2SiO4为模型矿物发现微生物铁载体能显著加快矿物的溶解速率。Barker等[18]研究表明生物膜内的pH较溶液的低,导致生物膜作用下长石的溶解速率增大了两个数量级。

1.3 矿物对细菌活性的影响

细菌吸附于矿物表面后,矿物通过其物理特性以及化学组成对细菌的生理活性造成影响[45]

矿物物理特性对细菌活性的影响主要表现为矿物与细菌吸附过程中矿物对微生物膜的破坏。Glasauer等[19]观察到纳米针铁矿可穿透细菌Shewanella putrefaciens外膜并刺穿肽聚糖层,与细菌表面结合,同时阻碍细菌表面养分吸收,抑制其活性。Cai等[20]发现细菌与蒙脱石、高岭石结合时活性未受影响,而与针铁矿结合时细菌的生物膜遭到严重破坏。Qu等[46]报道,赤铁矿颗粒在与细菌细胞接触后的10 h内,细胞膜结构完整性丧失,甚至可观察到颗粒迁移至细胞内部。Asadishad等[47]的研究中,细菌在与SiO2、Fe2O3,和Al2O3结合后会产生O-和C-金属键,使细胞膜受损而失活。此外,土壤矿物的粒径大小也会影响细菌的生理活性,但是目前关于矿物粒径对细菌活性影响的研究仍很缺乏。Zhou等[24]发现不同粒径大小(100 μm,100~50 μm,< 50 μm)的高岭石和蒙脱石均可加速Bacillus halophilus Y38的生长,并且矿物的粒径越小,促进作用越强。在矿物对细菌细胞膜完整性的影响研究中可使用Live/Dead BacLight Bacterial Viability Kits对活/死细胞菌体进行染色。该方法中包含SYTO-9和碘化丙啶两种染色剂。其中,SYTO-9染色通常能标记所有细菌,包括膜完好的和膜损坏的。而碘化丙啶只能穿透膜损坏的细菌。所以,在SYTO-9和碘化丙啶的混合染色中,细胞膜完整的细菌染色为荧光绿色(被认为是存活的),而细胞膜受损的细菌染色为荧光红色(被认为是死亡的)[20]

矿物化学组成对细菌活性的影响主要表现为矿物溶出的矿质离子对吸附的细菌造成毒害。目前相关研究多集中在溶出的Al和Fe离子对细菌活性的抑制。Zhou等[15]研究表明,细菌吸附于矿物表面后会破坏钙长石中的Al-O-Si键,释放出Al3+和Si3+。高岭石在中性pH条件下溶出的Al3+浓度为10-7~10-8 mol·L-1,而三水铝石Al(OH)3溶出的Al3+离子浓度为10-6 mol·L-1。同时,Wong等[26]研究发现高岭石、三水铝石等矿物中Al2O3含量与其对Desulfovibrio vulgaris活性的抑制作用存在较大相关性,并推断这种抑制作用是矿物中溶出的Al3+对细菌的直接影响。大量研究表明Al3+会毒害细菌,抑制细菌的活性。Flis等[48]发现当溶液pH为4.5和5.5时,5~50 μmol·L-1的Al3+即可抑制细菌生长。Guida等[49]研究同样表明Al3+的存在可抑制Escherichia coli的生长。在Al3+胁迫下细菌可通过调控相关基因的表达适应胁迫。Guzzo等[50]发现Al3+胁迫下Escherichia colifliC基因上调,fliC基因在细胞的存活响应中发挥着重要作用。此外,含铁矿物溶出的Fe2+和Fe3+也会对细菌细胞产生毒害效应,降低细菌活性。对于Fe2+而言,Williams等[51]认为,赤铁矿、针铁矿等溶出的Fe2+可进入细胞并被氧化成Fe3+沉淀,产生致命的氢氧自由基,使细菌失活。Park和Imlay[52]指出,当过氧化氢与Fe2+在细菌Escherichia coli体内发生反应形成氢氧自由基时,细胞DNA受到严重损害,细菌失活。细胞内羟基自由基的生成通常被认为是导致细胞死亡的潜在机制[53]。部分细菌可通过体内调节机制抵抗Fe2+的胁迫抑制作用。Moore和Helmann[54]指出,在Bacillus subtilis体内,PerR是感知Fe2+与过氧化氢反应的调节蛋白,具有抗氧化作用,体内的AhpC/F可通过分离和还原调控消除过氧化物,调节蛋白MrgA对Fe2+的隔离则可进一步保护细胞免受过氧化物的损害。而对于细菌Escherichia coli而言,细胞内的过氧化氢会激活其体内OxyR转录因子,直接刺激蛋白质的合成,其中的hemH基因在氧化胁迫下上调了11倍[55]。进一步的研究发现mntH是控制OxyR转录因子的基因之一,当Escherichia coli细胞受氧化胁迫时会激活mntH适应胁迫环境[56]。此外,溶出的Fe3+同样会对细菌产生毒害作用。Chamnongpol等[57]指出,Fe3+会损坏细菌外膜,并对细菌造成氧化胁迫。在这种氧化胁迫下,部分细菌可通过转录调节应对胁迫并存活。Yu和Ye[58]发现Bacillus subtilis在Fe3+胁迫下,全基因组中共有766个基因的表达被调控,其中有366个基因表达上调,400个基因表达下调。值得注意的是,Fe3+诱导的细菌氧化应激作用可引起SigB氧化抗性组基因sodFyqiGyqjM的表达上调,其中yqiGyqjM基因可调控细菌进行氧化应激排毒,以此适应Fe3+胁迫与毒害。而细菌Escherichia coli则通过提高由PmrA和QseB调控的qseBC基因的表达并激活PmrAB以应对高浓度的Fe3+[59]

此外,一些沉积在矿物表面的有毒金属元素、有毒有机物等同样会随着矿物的溶解而释放,并最终对细菌的活性造成毒害。但仍存在另一些观点认为矿物能促进细菌的活性。Rong等[21]的研究就提供了证据,其研究表明高岭土和蒙脱土刺激Bacillus thuringiensis细菌增长,增强其代谢活性。Chaerun等[22]研究同样发现矿物对微生物的生长存在促进作用。对于矿物促进微生物的生长一般存在四种假设:(1)矿物捕获环境中的代谢抑制剂,间接促进微生物生长;(2)矿物通过缓冲环境中的pH来维持微生物生长[23];(3)矿物作为电子受体支撑细菌的生长[25];(4)矿物作为微生物生长依附的支撑材料[21]

2 细菌-矿物复合体在重金属污染土壤中的应用

细菌-矿物复合体共同作用于土壤环境中的重金属,改变土壤中重金属元素的吸附、迁移行为,在重金属污染修复中具有重要意义。细菌与矿物作用形成复合体后,由于结合方式的不同可能导致复合体吸附能力的差异。方临川[60]探究了蒙脱石、针铁矿两种矿物与细菌的吸附行为,表明蒙脱石与Bacillus thuringiensisPseudomonas putida两种细菌形成的复合体吸附Cu2+的能力较理论值增加了16.4%和30.6%,而针铁矿与两种细菌形成的复合体吸附Cu2+的能力较理论值降低了19.6%和6.5%。自动电位滴定结果显示蒙脱石与两种细菌形成复合体后,表面位点浓度较预测的理论值增加了1.94%~6.20%。与之相反,针铁矿与细菌形成复合体后表面位点浓度却降低了2.57%~6.26%。研究者推测由于针铁矿-细菌间静电作用力强,两者的紧密结合覆盖掉表面一些重金属结合位点,使得吸附较理论值降低。而蒙脱石-细菌间相互作用较弱,松散的结合方式可能激活了复合体表面“新”的位点,故吸附较理论值增强。这一研究表明细菌-矿物结合方式的不同可能是影响复合体吸附能力的重要原因。对于结合后细菌-矿物复合体对重金属的吸附能力,目前研究尚存在两种有争议的观点。一方面,有些研究认为复合体的形成过程可能会掩盖部分细菌或矿物表面的官能团,使得复合体材料对重金属的吸附能力降低。Kulczycki等[61]研究了Bacillus subtilis-水铁矿复合体对Cd2+的吸附,结果表明复合体的实际吸附量低于根据组分相加原则预测的理论吸附量。Walker等[62]研究发现,Escherichia coliBacillus subtilis在与蛭石、高岭石形成复合体后吸附重金属Cu2+、Cd2+、Zn2+的量较细菌、矿物单一组分对重金属的吸附量减少20%~90%,表明细菌细胞壁表面的重金属吸附位点可能被掩盖。另一方面,一些研究却认为复合体的形成不但不占用重金属的吸附位点,反而能提高其对重金属的吸附能力。Zhu等[63]的研究发现针铁矿-Bacillus subtilis复合体吸附Cu2+和Cr3+的速率以及最大吸附量均较单一的针铁矿和细菌组分要高。Franzblau等[64]的研究同样发现,Anoxybacillus flavithermus在与氧化铁矿物形成复合体后吸附Cu2+的量较细菌、矿物单一组分的吸附量之和高出20%。Chen等[65]的研究表明,Pseudomonas putida与针铁矿的结合增强了针铁矿对Cu2+和Zn2+的吸附量,并未减少细菌表面的重金属结合位点。在Xu等[66]的研究中,黏土矿物和Cd2+的共同作用可促进细菌Serratia marcescens S14形成生物膜,最终增强对Cd2+的吸附。此外,复合体吸附重金属的特征与细菌种类、矿物类型、矿物/细菌质量比、环境因素等密切相关。Flemming等[67]的解吸实验表明黏土矿物-细菌复合体中重金属的解吸量随着解吸剂、细菌种类、矿物类型的不同而不同。研究者认为,由于不同解析剂、细菌种类以及不同矿物类型的表面特性不同,致使其对重金属的吸附、解吸特征不同。谢朝阳等[29]的研究表明,复合体中细菌组分的比例越大,吸附重金属的能力越强。而Du等[68]在对比例为10:1、5:1、2:1和1:1的蒙脱石/细菌复合体吸附重金属能力进行研究时同样发现当细菌的占比更高时,即矿物与细菌质量比为1:1时的复合体吸附重金属能力最强。Qu等[69]观察到当体系中pH > 5.5时,在Cu2+的作用下可形成蒙脱石-Cu-假单胞菌桥架结构,促使复合体对Cu2+的吸附量增加;而体系pH < 5.5时,由于蒙脱石与假单胞菌间存在表面位点的掩盖情况,导致复合体吸附Cu2+的能力下降。此外,随着离子强度的增加(0.1~0.01 mol·L-1 NaNO3),复合体对Pb2+的吸附能力也相应下降。其中质量比为12:1的蒙脱石/假单胞菌复合体对Pb2+的吸附量降幅约为15%,相比之下,在2:1复合体上则仅观察到8%的降幅。研究者认为这一现象表明复合体对Pb2+的吸附以内层络合为主[70]

由于复合体中细菌与矿物组分对重金属存在竞争吸附,使重金属在复合体上的分配和迁移出现差异。目前许多研究表明复合体的细菌组分对重金属的固定起主要作用。Walker等[62]在Ag+,Cu2+,Cd2+,Ni2+,Pb2+,Zn2+,Cr3+硝酸溶液中研究了E. coli K-12细胞外膜、Bacillus subtilis 168细胞壁与蒙脱石和高岭石的相互反应,发现复合体对重金属的吸附能力由高到低依次为:细胞壁-蒙脱石、细胞壁-高岭石、细胞外膜-蒙脱石、细胞外膜-高岭石。通过能谱分析结果显示,细菌是复合体中固定重金属的主要组分,各组分吸附重金属的能力由高到低依次为:细胞壁、细胞外膜、蒙脱石、高岭石。研究者认为单组分的金属结合能力由其净电负性以及形成静电和化学键的能力共同决定,并最终反映在其阳离子交换能力上。其中,枯草芽胞杆菌的细胞壁较大肠杆菌的外膜结合金属的能力更强,这是由于革兰氏阳性细胞壁中肽聚糖的数量和电荷密度更大,而肽聚糖负责大部分的金属结合。虽然细胞外膜的表面也含有很多带电基团,但这些基团通常被多价金属离子中和而表现出较弱的金属结合能力。相较于高岭石而言,蒙脱石具有更高的阳离子交换能力,故而吸附重金属的能力较强。Chen等[71]发现假单胞菌-针铁矿复合体较菌体固定重金属的能力低,其中复合体的细菌组分对Zn2+的固定作用最强。Du等[72]同样发现Cd2+更倾向于固定在恶臭假单胞菌-蒙脱石复合体中的细菌表面。而Qu等[70]却发现细菌的存在可促进Pb2+与蒙脱石形成桥架结构,这种桥架结构最终可增加Pb2+在矿物组分上的分布。

虽然近年来越来越多的研究探索了复合体对重金属的固定作用,为复合体应用于土壤重金属污染修复提供了一定的科学依据,但是由于土壤以及复合体结合过程中两组分间相互作用的复杂性,细菌在与土壤矿物结合成复合体后对重金属吸附能力的影响及作用机制仍有待进一步的研究与实践。

3 展望

在土壤修复中,除重金属胁迫外,土壤矿物也会对细菌定殖及定殖后固定重金属的能力造成影响。目前关于土壤矿物-细菌-重金属三者之间的研究仍很缺乏,且大多集中在表观现象层面。因此,在前人研究空缺基础上可从以下几方面展开更深入的研究,揭示不同矿物影响下微生物的分子响应和生理调控与固定重金属机制的内在联系,明晰土壤矿物-微生物-重金属三者间的相互作用,为微生物-植物联合修复应用于重金属污染土壤环境提供科学依据。

1)在分子水平上综合分析不同表面特性的矿物与微生物之间结合特征与结合方式的差异,并深入探究这种差异对重金属在微生物-矿物二元系统表面的分配与固定造成的影响。土壤矿物和微生物的表面特性与结合方式不同可在一定程度上影响两者表面的官能团类型、浓度和电荷分布等,那么不同复合体的结合特征最终如何影响重金属的固定与分配?结合后的复合体较单一组分而言固定重金属的机制有何不同?

2)深入研究不同矿物对微生物定殖、生理活性、代谢活动、抗氧化系统等的影响以及在此影响下微生物的分子响应与调控机制,并进一步探究由此对重金属固定产生的影响与作用机制。例如微生物与矿物结合可能会破坏微生物的细胞膜,溶出的矿质离子也可能对微生物造成毒害,诱导羟基自由基释放,致使微生物代谢紊乱,进而影响一系列的微生物过程。那么在这种胁迫下微生物体内的分子响应和调控机制是怎样的?最终是否会对微生物在土壤环境中定殖以及固定重金属的作用造成影响?

3)进一步明晰重金属与矿物共同存在/影响下微生物的响应调控机制(如相关抗性基因的表达与转录,代谢产物量与类型的变化等)。大量研究表明,在重金属胁迫环境下微生物可通过一系列的适应机制而存活,那么当矿物与重金属共存时,微生物受到的胁迫是增强亦或是由于矿物对重金属的吸附作用反而可缓解部分重金属对微生物造成的胁迫?与单一重金属胁迫或者矿物影响相比较,微生物在矿物与重金属共存环境下的适应机制有何不同?
